Thanks for the suggestions. However, I already have two metabolites in the model and 10-12 covariates with only 1- 4 entries per subject. I think after considering wt, age, type covariates for inclusion in the model I'll look at the DDI 'covariates' post Pmetric analysis.

I'm trying to use covariates as a way of including drug interactions on the primary drug parameters. However, not all subjects receive the same potentially interfering drugs. Thus, there are many missing entries for the secondary drug exposure. Entering 0.0 allows the analysis to proceed as does -99...

Oh dear. It looks like I had some extra empty columns in the .csv file. I had them in my overall spreadsheet for extra covariates and exported the data as the .csv file for Pmetrics. Deleting the unnecessary, empty columns seems to have fix the 'problem'.

Hmm. I've got three test runs working. Drug, Drug and one metabolite and Drug with two metabolites. Randomly generated data. You can have different number of parameters for different models but the error functions should be the same.

There are various criteria for choosing the 'best' model. Residual plots, AIC, -2*LL, parameter cv, etc. Sometimes they all point in the same direction.
